en el evento cellendedit del dgv puse esto
Dim total As Single
Dim precio As Single = dvg_tienda.Item(2, dvg_tienda.CurrentRow.Index).Value
Dim cantidad As Integer = dvg_tienda.Item(3, dvg_tienda.CurrentRow.Index).Value
total = precio * cantidad
dvg_tienda.Item(4, dvg_tienda.CurrentRow.Index).Value = total
